- All-Star Cast: LEGO DC Super-Villains features a lot of the same talent as the previous game, but also has names like John Barrowman, Michael Ironside, Michael Dorn, and Mark Hamill all reprising their roles, and several more actors that were from the DCAU, like Kevin Conroy, Michael Rosenbaum, and Susan Eisenberg.
- Awesome, Dear Boy: Aside from So My Kids Can Watch, Stephen Amell confirmed that this is one reason he returned to voice Green Arrow.

- Milestone Celebration: There is a DLC Pack for LEGO Batman 3: Beyond Gotham that features different skins for Batman and some of his various supporting characters from the 75 years he's been around.
- The Other Darrin: A good chunk of the voice cast changes for LEGO Batman 3.
- Aquaman is voiced by Scott Porter, instead of Brian Bloom. Similarly, Bumper Robinson plays Cyborg again, taking Bloom's place.
- Josh Keaton takes over as Green Lantern and Nightwing from Cam Clarke and also takes over the role of Shazam from Travis Willingham.
- Dee Bradley Baker takes over from Troy Baker as Braniac, and Marc Worden also takes Troy's place as Sinestro. Troy in turn takes over from Joseph Balderrama as Killer Moth, and also replaces Nolan North as Hush.
- Laura Bailey takes over as Catwoman from Katherine Von Till. Grey Griffin takes over in Lego DC Supervillains.
- Rob Paulsen is replaced by Roger Craig Smith as the Riddler. Wally Wingert takes over for LEGO DC Supervillains.
- In Lego Batman 2, Steve Blum voiced Bane, but in the third game, JB Blancreprised the role.
- Christopher Corey Smith, who had played The Joker since LEGO Batman 2, was replaced by Mark Hamill for LEGO DC Supervillains.
- Troy Baker was replaced with Kevin Conroy for Batman in LEGO DC Supervillains.
- Laura Bailey was replaced by Susan Eisenberg for Wonder Woman in LEGO DC Supervillains.

- Playing Against Type: In the first game, Steve Blum portrays Batman, Killer Croc (a role he would go on to play without the Simlish in Batman: Arkham Asylum and Batman: Arkham City), the Joker, Two-Face, and, of all characters, Killer Moth.
- Real Song Theme Tune: Wolfmother's 'Joker & the Thief' was heard in the main menu of DC Super-Villains.
- Refitted for Sequel: Darkseid was planned to be playable in LEGO Batman 2, but was dropped, with the only hints left being unusable character data. He was in the final batch of DLC characters in LEGO Batman 3.
- Show Accuracy/Toy Accuracy: DC Super-Villains notably veers away from toy accuracy and toward show accuracy, a rarity for the LEGO games. Of the characters who have real-world minifigs, very few actually resemble the toys.
- So My Kids Can Watch: In this interview, Stephen Amell admitted that one of the main reasons for his reprising Green Arrow was so his young daughter could see something that he was in. Awesome, Dear Boy factored into it too.
- Trolling Creator:
- Due to fan demand for Raven for Beyond Gotham, the official Twitter account started teasing fans about it. Firstly, they jokingly asked if she was even that popular, then pretended to confuse her for Raven-Symoné by tweeting '#RevealRavenSymone'. Finally they agreed to reveal Ra..'s Al Ghul.
- One of the Post-Credits sequences in LEGO Batman 3 features a silhouetted group shot that appear◊ to be another company's iconic characters. Only it turns out they're Black Canary, Aquaman, Nightwing, Swamp Thing, Shazam!, Booster Gold, and Green Arrow.
- What Could Have Been:
- According to Brickipedia, the Lord of Apokolips himself, Darkseid, has character data that can be found by digging through the game files in LEGO Batman 2: DC Super Heroes. Darkseid is a playable character, however, in DLC for LEGO Batman 3.
- Nora Fries, the cryogenically frozen wife of Mr. Freeze, was also planned, but possibly scrapped due to lack of importance and canonically not making sense as a playable character.
